In Vitro Anticancer Effect of Chungkukjangs from Folk Villages of Sunchang Region in HT-29 Human Colon Cancer Cells

Abstract
Six different chungkukjang products (K-, M-, Mn-, O-, Os-, H-chungkukjangs) were purchased from Folk Villages of Sunchang region, and 3 samples, K-, H- and M-chungkukjangs showed higher sensory evaluation and good fermented quality among the samples. The three chungkukjang samples showed in vitro anticancer activities in HT-29 human colon cancer cells. The anticancer effect of the 3 samples all showed same trends, however K-chungkukjang exhibited the highest anticancer activities in the HT-29 cells. MTT assay, growth inhibition tests and apoptosis analysis by DAPI staining and RT-PCR employed to check the anticancer effect in the colon cancer cells. At the concentration of 1.0 mg/ml, K-chungkukjang (86%) showed the highest growth inhibitory effect in the cancer cells, followed by H-chungkukjang (76%) and M-chungkukjang (44%) (p£¼0.05). K-chungkukjang induced apoptosis as determined by DAPI staining and exhibited the increased Bax and decreased Bcl-2 in mRNA expression. Among the chungkukjang samples, K-chungkukjang showed the highest anticancer effects. It is concluded that chungkukjangs revealed anticancer activities, but the activity exhibited differently depending on the different product, probably due to the preparation methods.
